Simple Steps for Your Tissue Culture Nursery
Step 1: Giving the Time to Adjust
When you receive your tissue culture plantlets, do nothing for the first 1-2 days to ensure they are adapted to their new temperature.
Step 2: Applying Fungicide and Mixed Water
To mitigate the risk of fungal infections, it is advisable to prepare fungicides such as Captain, carbendazim, or betadine. These should only be utilized for 15 minutes, in conjunction with a rooting hormone. Subsequently, pour the fungicide and water mixture onto the planting material. It is crucial to exercise caution and avoid overwatering, ensuring the soil remains consistently moist.
Step 3: Bringing Plantlets Out
Bring culture plantlets out of the plastic bag, and remember to trim away anything that needs to be removed by clearing anything that might obstruct the plantlets' growth. After the clearing process, put the plantlets into the water.
Step 4: Cleaning the Root and Rest
Use a gentle brush to clean any remaining gel from the roots until they are entirely clean and do not have residue. Following the cleaning process, immerse the plantlets in mixed water for a duration of 3 to 5 minutes.
Step 5: Drying and Planting
Thoroughly dry the plantlets' tissue before planting them in the pot, ensuring that they are buried at least 2 inches deep or that no roots are exposed above the soil.
Step 6: Before the Last Step to Grow
Place the potted plant into an airtight container for 3 to 4 weeks, ensuring that the container is tightly sealed. Before sealing, generously water the plantlets and allow any excess water to drain off. During this phase, it is advisable to keep the plantlets in a shaded area and shield them from direct or intense sunlight.
Step 7: Slowly Introduce to Fresh Air
Please ensure that the plantlets remain covered for the initial seven days. Subsequently, over the next 7 to 10 days, gradually accustom to ambient air by allowing an hour of natural light and air every few days. Slowly increase the duration until the plantlets are fully acclimatized to their new surroundings.
Recommendation
Please adhere to the following guidelines for plantlets care:
1.) Use a mixture of peat moss and perlite in a 3:1 ratio.
2.) Do not use worm castings or fertilizers.
3.) Do not use hydrogen peroxide as a substitute for fungicide.
4.) Do not open the container unless the plantlets show signs of dehydration.
If necessary, carefully moisten the soil, ensuring the water does
not touch the plantlets.
If there’s too much condensation on the dome, you can open it for up to 10
seconds to vent.
5.) After 2-3 days, check the container. If there is no condensation inside,
add moisture to sustain the plantlets.
6.) Label the container with the plant's name and planting date
for proper record-keeping.
7.) Maintain humidity levels between 80% and 99% for optimal growth.
8.) Provide proper lighting, preferably using an LED tube T8.
